Despite recent advances, there remain significant needs in the treatment of solid tumors, which make up 90% of all cancers. While recent treatment innovations, including immunotherapies, have improved patient care, the CDC reports<\/a><\/span> that more than 600,000 people die of cancer in the U.S. every year. The constant evolution and highly personalized nature of cancer cells and their remarkable ability to impede a patient\u2019s immune response continue to challenge currently available therapies’ ability to selectively kill them, whether by targeted therapies or by engaging the immune system.<\/p>\n

We believe our CRISPRomics platform has generated the genomic insights that will enable us to overcome this challenge.<\/p>\n

By their very nature, Tumor-Infiltrating Lymphocytes (TIL) are an ideal therapy for the treatment of solid tumors, as they are a patient\u2019s personalized immune response against cancer cells and possess broad, polyclonal T cell receptor-mediated recognition of cancer cells. This polyclonality provides TIL with the potential to eliminate every last cancer cell, even if the cancer cell evolves. In fact, the first TIL therapy has now been approved in the U.S. for the treatment of melanoma. Despite this promise, however, for many patients, TIL therapy unfortunately falls short.<\/p>\n

So, how does our approach improve upon this?<\/strong><\/span><\/p>\n

We seek to maximize the tumor-killing activity of TIL therapy<\/strong> by knocking out our top CRISPRomics-discovered targets, SOCS1<\/strong> and Regnase-1<\/strong>, in TIL using CRISPR\/Cas9 gene editing creating engineered Tumor Infiltrating Lymphocyte, or eTIL\u00ae<\/sup> therapies. In our animal models to date, inactivation of Regnase-1<\/strong>\u00a0and SOCS1<\/strong> has demonstrated a remarkable ability to boost the ability of T cells to infiltrate and kill tumors, even in settings where no other therapies work.<\/p>\n

By engineering into each TIL best-in-biology anti-tumor activity, we believe eTIL therapies <\/span>may solve the intractable efficacy obstacles facing patients currently receiving T cell therapies in the treatment of solid tumors.<\/p>\n

We believe our <\/span>eTIL programs <\/span>may be a solution to <\/span>the Solid Tumor Challenge.<\/strong><\/span><\/p>\n

Dr. Rizvi has a record of success in building exceptional teams that deliver results. Prior to joining KSQ, Micah served as the Head of Tolerance Therapeutics in the Immunology and Inflammation Research Unit at Pfizer, Inc., where he was responsible for building and leading a drug discovery portfolio and identifying external research opportunities focused on immune tolerance induction therapeutics. He earned his PhD in Immunology from Dartmouth Medical School where his work on B cell survival and T regulatory cell differentiation generated fundamental insights into the mechanisms of immune-mediated diseases. From 2013 to 2017, prior to joining PhRMA, he served as the Deputy Center Director for Science Operations for the U.S. Food and Drug Administration\u2019s (FDA) Center for Drug Evaluation and Research (CDER). Rich was senior vice president and head of Clinical Development at Sanofi-Genzyme from 2011-2013. He was Chief Medical Officer at Genzyme Corporation from 1992 to 2011, where he was responsible for worldwide global regulatory and pharmacovigilance, as well as all aspects of clinical research and medical affairs.<\/p>\r\n\r\n

Rich is board certified in internal medicine, diagnostic and laboratory immunology, and allergy and immunology. Prior to joining KSQ Therapeutics, Frank served as Director and Head of Global Oncology Target Discovery at the Novartis Institutes for Biomedical Research (NIBR), where he was responsible for developing the early oncology drug discovery portfolio. Frank also led the therapeutic siRNA research strategy at Novartis and directed the Cancer Cell Line Encyclopedia collaboration with the Broad institute. During his tenure at Novartis, Frank and his team identified more than a dozen novel oncology targets and advanced multiple chemical entities addressing these targets into preclinical development. He also led the biology efforts to advance the PKC inhibitor Sotrastaurin into phase I clinical trials in lymphomas and uveal melanoma.
